چکیده

Prothrombin is activated by activated Factor X f (Factor V and phospholipid) in the presence of diisopropylfluorophosphate which preferentially inhibits thrombin. Under these conditions a single activation fragment, designated Fragment 1.2, is released which represents the complete nonthrombin portion of the prothrombin molecule. The same fragment is seen as a transient species in the absence of diisopropylfluorophosphate when the activation reaction is monitored by disc electrophoresis in 8 M urea. The NH2terminal residue of both prothrombin and Fragment 1.2 is alanine, suggesting that Fragment 1.2 is derived from the NH&erminal end of the prothrombin molecule. Fragment 1.2 is not cleaved by activated Factor X f (Factor V and phospholipid), but is cleaved by thrombin to form two fragments which are identical by electrophoresis, ion exchange chromatography, and amino acid composition to Fragment 1, the fragment released from prothrombin on incubation with thrombin, and Fragment 2, the fragment released by activated Factor X when Intermediate 1 is converted to Intermediate 2 (Paper I; OWEN, W. G., ESMON, C. T., AND JACKSON, C. M. (1974) /. Bi02. Chem. 249, 594-605). It is proposed that the formation of Intermediate 1 during prothrombin activation is solely the result of thrombin-catalyzed proteolysis of prothrombin.

Formation of meizothrombin as intermediate in factor Xa-catalyzed prothrombin activation.

The conversion of prothrombin into thrombin by Factor Xa requires the cleavage of two peptide bonds in prothrombin. Dependent on the order of cleavage, prethrombin 2 or meizothrombin occurs as intermediate. Since prethrombin 2 has as yet been the only observed intermediate, prothrombin activation is generally considered to proceed via prethrombin 2. In this paper we present new methods that all...

Crystal structures of prethrombin-2 reveal alternative conformations under identical solution conditions and the mechanism of zymogen activation.

Prethrombin-2 is the immediate zymogen precursor of the clotting enzyme thrombin, which is generated upon cleavage at R15 and separation of the A chain and catalytic B chain. The X-ray structure of prethrombin-2 determined in the free form at 1.9 Å resolution shows the 215-217 segment collapsed into the active site and occluding 49% of the volume available for substrate binding. Remarkably, som...

Role of prothrombin fragment 1 in the pathway of regulatory exosite I formation during conversion of human prothrombin to thrombin.

Prothrombin (Pro) activation by factor Xa generates the thrombin catalytic site and exosites I and II. The role of fragment 1 (F1) in the pathway of exosite I expression during Pro activation was characterized in equilibrium binding studies using hirudin(54-65) labeled with 6-(N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino)hexanoate ([NBD]Hir(54-65)(SO3-)) or 5-(carboxy)fluorescein ([5F]Hir(54-65)(...

Effects of activation peptide bond cleavage and fragment 2 interactions on the pathway of exosite I expression during activation of human prethrombin 1 to thrombin.

Activation of prothrombin (Pro) by factor Xa to form thrombin occurs by proteolysis of Arg271-Thr272 and Arg320-Ile321, resulting in expression of regulatory exosites I and II. Cleavage of Pro by thrombin liberates fragment 1 and generates the zymogen analog, prethrombin 1 (Pre 1). The properties of exosite I on Pre 1 and its factor Xa activation intermediates were characterized in spectroscopi...

Expression and Characterization of Recombinant Ecarin

The snake venom protease ecarin from Echis carinatus was expressed in stable transfected CHO-S cells grown in animal component free cell culture medium. Recombinant ecarin (r-ecarin) was secreted from the suspension adapted Chinese Hamster Ovary (CHO-S) host cells as a pro-protein and activation to the mature form of r-ecarin occurred spontaneously during continued incubation of the cell cultur...

Recombinant Factors for Hemostasis

Thrombin is the key serine protease in hemostasis because it is responsible for activating both procoagulant proteins critical for creating fibrin clots and anticoagulant proteins needed to return the system to balance. Prothrombin is converted to thrombin in primary hemostasis by activated Factor X (FXa) attached to the phospholipid membrane and secondary hemostasis by the prothrombinase compl...
